as for the OP, it's been a long time since I've watched Blood+ but if as I recall

I don't think the "only Diva's blood can kill him" argument is 100% solid.   While it makes sense, when he and Solomon are fighting at one point,  Solomon drags him high up into the sky and comments that even a Chavalier could not survive a fall from that height.

As for how they got to James,  the short answer is they didn't.  At least that is how I interpret it.  The whole project was based on genetic manipulation, so it makes sense they would have DNA from everyone on file for whatever reason.   I believe that the James at the end of the series was just a clone,  and I point to the episode at Nathan's house where he comments on James sleeping / needing his sleep / whatever and it cuts to a shot of what appears to be a tank with a "small" version of James in it.   It could be postulated that they grew a new clone and even if his memories did not carry over, and he was a simple human clone,  he would simply need to drink Diva's blood.   This is also supposing that the memory carrying effect of blood goes both ways and not just from Chavalier to newly awakened Queen.

Coincidentally this could also explain how Nathan remained alive at the end of the series..   I don't know where the train of thought that he was not one of Diva's Chivalier comes from,  so I can't speculate on that.  However  he is clearly a gay ass drama queen and is not the type that would want to  die and not learn how things turned out,  or not have a backup plan  in case he was killed for some unforeseen reason.
