It may be that she has a scar there or that it's to help identify her.

The identification we already have with the pig tail.
Looks more in the moe realm than anything else. Maybe the manga will explain the reason someday...

I'm fine with it being just some sort of beauty mark. Regardless, it doesn't make me love Riko any less, even if she weren't wearing one.

Dialogue goes then like this:
*Flashback*
Keita: "But if you can't become a bride because of that scar, then..."
Riko: "What are you saying?"
Keita: "Eh..?"
Riko: "Aren't you the one who is going to make me a bride, Keita?"
*End Flashback*
Ako: "And so ... apparently that's why even now, by hiding that scar, she's praying the day she gets to marry Kei-chan would come"
